The event is a partnership between English Schools' Football Association, Plymouth Schools FA and Plymouth Argyle and will be the second game for the England Schoolboys in this season's Centenary Shield.
By the time they reach Plymouth, England U18s will have played Wales at Lincoln City before two away fixtures in April against Scotland and the Republic of Ireland to complete the competition.
England will be hoping to defend their current status as champions during the 2012/13 campaign after winning the Shield in the final game of last season's competition.
Chris French, Plymouth Schools FA chairman, believes the international will excite and attract fans of all ages.
He said: "This is an fantastic opportunity for the city to host an England international at Home Park.
"We are looking for this event to involve the whole city, from children of all ages coming to watch to companies who want to be part of the event.
"Maybe they'll see the stars of the future."
